Significant Facts Two Utah sisters, Maria and Jan Brady posted a message on Facebook in support of anti-globalization and anti-genetically organism (GMO) activists against McDonalds restaurants, who had incited riots in Europe. The sister’s message, sent to over 500 â€Å"friends†, asking them to join the sisters in two days at 10 p.m. at a McDonalds near the sister’s apartment for a â€Å"night of riot, pillage, and fun†. A secondary message was sent out asking the participants to bring items that could be used as weapons or cause property damage. However, one of the â€Å"friends† that the message was sent to turned the postings over to the Salt Lake Police Department. â€Å"Mother to Son,† written by Langston Hughes, is a short poem about a mother who is teaching her son about perseverance and determination by using the image of a staircase. She explains that even though life has given her many hardships, she continues forward and she urges her son to do the same. In â€Å"Mother to Son,† Langston Hughes uses an extended metaphor, imagery, dialect, and structure to paint a picture of a weary mother who wants her son to persevere through the hardships of life. The theme of this poem is determination and the value of experience. The mother is telling her son that no matter what happens, he should never give up. There are many different life forms on earth. New species and different sub species are evolving every second as others die out. In order to explain the origin of life, people have used the bible story of creation, while others came up with their own theories. Out of all those theories, the theory of evolution by Charles Darwin seems to have the most supporting evidence to back it up. Evidence on earth suggests that single cell creatures are the ancestors of all life. These organisms slowly evolved into air breathing creatures when oxygen was created. Slowly, the as land dried up, these creatures moved away from the water and on to land where they evolved legs and a strong back bone. The theory of evolution by Charles Darwin explained how life evolved from survival of the fittest, it provides a logical explanation of the evolutionary process these creatures took, and how human came to be. Evidence on earth shows that single cell organisms eventually evolved to multicelled organisms and complex beings. If you look at old fossil records, the oldest fossils are all single cell creatures. These simple creatures do not require oxygen to live. It took in energy from the sun and other energy sources. Slowly as oxygen begins to appear on Earth, some of the organisms began to evolve lungs to breathe oxygen because When free oxygen entered the picture. Breathing is more efficient than drawing energy from the fermentation process that earlier microscope life used. This dye binds mostly to arginine, tryptophan, tyrosine, histidine, and phenylalanine residues of the protein Materials and Reagents Protein standard: 1mg/mL Albumin Bradford reagent Distilled water Test samples A, B and C (Unknown protein) One 96-well plate Procedure First, the albumin standard solutions were prepared as follows: Concentration Albumin Distilled Water 0% 0ul 100ul 25% 25ul 75ul 50% 50ul 50ul 75% 75ul 25ul 100% 100ul 0ul Then the Bradford reagents were diluted with 300 ul (Bradford): 1500ul (Distill Water) (1:5 ratio). In the first trial, 180ul of the diluted Bradford reagents was added into the 96 well plate. Then 0%, 25%, 50%, 75%, 100% and Sample A, B, C, each 5ul was added to each well hole which contained 180ul of diluted Bradford reagent. These were tested with spectrophotom eter and the results recorded. The experiment was repeated again the same way. In addition, Sample C was diluted with 3 different ratios as follows: 1:9 (Sample C: Distill Water) 1:99 (Sample C: Distill Water) 5:95 (Sample C: Distill Water) The results were recorded after the solutions were tested with a spectrophotometer. A standard curve of absorbance versus concentration protein was drawn. Absorbance values of unknown samples were then interpolated onto the plot for the standard curve to determine their concentrations as shown in the graph below. Discussion From the graph abov e, the optimum measurement wavelength for this assay is on sample C. Thus sample C has the highest protein concentration.
